Sphingolipids and their metabolizing enzymes are key mediators in vivo. Sphingosine kinases (SphKs) and their lipid product sphingosine 1-phosphate (S1P) play an important role in signaling and in the development and progression of diseases, especially in chronic inflammatory and autoimmune diseases. S1P is a sphingolipid metabolite with multiple biological activities and is widely present in platelets, erythrocytes, neutrophils and lymphatic fluid, blood and other cells and body fluids.S1P is generated and can act intra- and extracellularly. On the one hand, it acts on cell surface receptors through autocrine and paracrine modes to exert its extracellular effects, including cell proliferation and migration, inflammatory cytokine infiltration, angiogenesis, and autoimmunity. On the other hand, it functions as an intracellular second messenger and acts directly on intracellular targets to regulate Ca2+ levels, gene transcription, protein modification, etc.
