Jasmonic acids (JA) are a class of lipidic plant hormones, synthesized from linolenic acid present in the chloroplast membrane. They are involved in development, abiotic stress responses and plant-microbes interactions in defence and symbiosis.
In abiotic stress, JA is usually involved in physiological and molecular responses. Physiological responses often include activation of the antioxidant system (superoxide anion radical, peroxidase, NADPH-oxidase), accumulation of amino acids (isoleucine and methionine) and soluble sugars, and regulation of stomatal opening and closing.

JA are also involved in a huge number of plants expanding events, such as primary root growth, leaf senescence, and reproductive development. Moreover, JA is implicated into the regulation of various metabolites production, such as phytoalexins and terpenoids.
Currently, Creative Proteomics presents an LC-MS/MS method for fast and highly sensitive determination of JA. A single-step sample preparation procedure based on mixed-mode solid phase extraction was efficiently combined with essential improvements in mobile phase composition yielding higher efficiency of chromatographic separation and MS-sensitivity. This strategy resulted in dramatic increase in overall sensitivity, allowing successful determination of phytohormones in small (less than 50 mg of fresh weight) tissue samples. The method was completely validated in terms of analyte recovery, sensitivity, linearity and precision.
