A globoside is a type of glycosphingolipid with more than one sugar as the side chain (or R group) of ceramide. The sugars are usually a combination of N-acetylgalactosamine, D-glucose or D-galactose. Recent research shows that globoside has many biological functions. Gb3 globosides specifically interact with the cellular "death receptor" FasR. When paired to a Gb3, FasR transduces a cell death signal derived from activation of the caspase-8 cascade. A genetic defect in the enzyme alpha-galactosidase leads to accumulation of Gb3, causing a lysosomal storage disease. Further, Gb3 are a component of the cell membrane where they bind to Shigatoxins facilitating their entry into cells. Gb4 glycoside is the main glycolipid found in the cell membrane of human red blood cells. There, they are essential for cell interactions including blood type determinants, which determine the adaptability of donor blood to patients. Each of the three different blood group antigens is defined by the variation of the oligosaccharide unit. Gb4 lipid also acts as a receptor for parvovirus B19, which infects rapidly dividing intestinal cells and bone marrow, causing children.
